Replication-competent virus (RCV) testing
Lentiviral vector-transduced cells should be used at Biosafety level 2. However, several institutes allow in vivo experiments to be downgraded to Biosafety level 1 if the cell have been tested and found negative for replication-competent virus (RCV). Imanis offers RCV testing for lentivirus using an enzyme-linked immunosorbent assay (ELISA) directed against the lentiviral p24 capsid protein. This rapid and highly-sensitive assay is the most widely-accepted test for detecting RCV. The limit of detection for our assay is 12.5 pg/mL of p24.
The cost for RCV testing is $425 per sample with bulk discounts available for order of 5 or more samples. The negative control is included in the price and is not counted as an additional sample.
Samples should be submitted as culture supernatants from cells grown for at least two weeks after lentiviral vector transduction. Culture supernatants collected earlier may yield false positive results. A negative (media only) control should also be submitted for each sample being tested.
Lentivirus titration by qPCR
Having an accurate titer for lentiviral vector stocks is important for optimizing transductions. Imanis can titer your lentiviral vectors using the same precise quantitative PCR (qPCR)-based method used for our extensive catalog of lentiviral vectors. Unlike traditional p24 ELISAs that fail to distinguish between functional and defective lentiviral particles, qPCR of integrated lentiviral genome copies exclusively measures the number of functional transducing units (TU/mL), giving you a more accurate titer to begin your studies.
Our standard qPCR titration targets the WPRE, a post-transcriptional regulatory sequence commonly included in lentiviral vectors. For titration of lentiviral vectors that do not contain a WPRE, please refer to our contract service page.
