How it Works

The Cellective platform is a two-component system for the manufacturing of lentiviral vectors (LVs) targeted to specific cellular receptors. Combining a highly optimized programmable G-fusogen with covalently bound targeting adapters, Cellective is easily incorporated into standard LV workflows.

Benefits of the Cellective Lentiviral Vector Delivery Platform

Cellective enables precision transduction of target cells, reducing biological noise, simplifying the study of target cells, and overcoming hurdles with co-culture models.

Cellective LVs encoding GFP and labeled with the CD3 Targeting Adapter efficiently transduce CD3+ Jurkat cells with minimal transduction of CD3- Jurkat-TCR-KO cells compared to traditional VSV-G pseudotyped LVs. 

Cellective makes it easier to transduce hard-to-transduce cell lines, including primary immune cells (PBMCs).

Unactivated PBMCs were efficiently transduced with Cellective LVs targeted with the CD3 Targeting Adapter, but not with untargeted Cellective LV control. Transduction efficiency with CD3-targeted Cellective LV was 3-fold higher than with a traditional VSV-G LV. 

Cellective CD3-retargeted LVs work in vivo to target and transduce T cells.

NSG DKO mice bearing BCMA+ OPM2-luciferase tumors were humanized with PBMCs. One week later (Day 0), mice were injected via tail vein with saline control, untargeted Cellective LV control, CD3-targeted Cellective LV, or Cellective Targeting Adapter only. Tumor burden was monitored over time by bioluminescence imaging. 

The Cellective Workflow

For both in vitro and in vivo applications, reach your intended target with Cellective. Here’s how:

Step 1: Transfect Cells

Transfect cells (e.g., HEK-293T) with Cellective LV Packaging Mix and desired lentiviral transfer plasmid. The packaging mix includes tat and is compatible with second- and third-generation transfer plasmids.

Step 2: Collect Particles

Collect particles containing the programmable G-fusogen. Concentrate or purify as needed for downstream application. Quantitate particle concentration by p24 ELISA.

Step 3: Combine Particles

Combine particles with the Cellective Targeting Adapter specific for your target of interest. Our ready-to-use Cellective Targeting Adapter Library currently includes adapters for targeting human CD3, with more in development.
