Quality and performance start before development. With deep scientific knowledge, our team will work with you to adjust product parameters based on our experience successfully working with different cell lines and applications. If we think there is a better way to meet your research needs, we will discuss it with you. By taking time to fully understand what you need to accomplish with your custom cell line, we are able to identify key performance characteristics for the products and test them during the development and QC process.
Custom cell line products are an ideal solution when the exact combination of tumor cell line and modification isn’t available as a standard product yet. Most frequently, this happens when you have a tumor model that you have established for studies, but you need it modified to support a new project direction or develop a different therapy.
Imanis has several established methods for cell engineering. For transgene overexpression, we primarily use lentiviral vector transduction. We have a ready-to-use in-house library of lentiviral vectors encoding reporter genes and TAAs of interest, as well as lentiviral plasmids that can be used to quickly clone in new reporters of interest as needed for projects. For knock-out/knock-down, we rely on CRISPR. While not part of our typical workflows, we also have the capabilities to engineer knock-ins using CRISPR or PiggyBac to facilitate more user-specific requirements.

Most of the time, monoclonal cell lines are considered superior, but polyclonal cell lines can have advantages for certain applications. Polyclonal cell lines represent more of the diversity of responses typically associated with a tumor. However, this diversity can also make experiments challenging, and monoclonal lines are often preferred due to their more consistent performance between experiments. In vivo, while clones generally exhibit more reproducible growth and responses, finding the right clone for a particular application can require screening multiple clones in vivo, as in vitro growth does not always correlate to in vivo growth. Monoclonal cell lines also take longer to engineer, which can be a factor to consider if fast turnaround is needed. However, engineered polyclonal cell lines must express a selection marker to ensure the modification is present on all cells of the population, and therefore not compatible with knock-out/knock-down modifications. For overexpression lines, for ultimate flexibility, we can provide both polyclonal and monoclonal cell lines. Polyclonal cells can ship earlier to facilitate early experiments while monoclonal cell lines are being developed. For monoclonal cell lines, we generally provide 2-3 clones for each modification to facilitate clone screening and comparisons, and can even tailor clones to express various levels of transgene if requested.
