Harnessing the Power of CD3-Targeted LVs

Selectively targeting LVs to CD3 using Imanis LV targeting reagents offers several advantages:

How the Cellective and G-Link Platforms Work

Imanis offers reagents for two LV retargeting platforms: Cellective and G-Link. Both technologies redirect VSV-G to CD3 using a protein adapter but achieve targeting through slightly different mechanisms.

Blinding
The VSV-G glycoprotein used to pseudotype the LV is “blinded” to its natural receptor, LDLR to prevent transduction of non-target cells.
Cellective uses a modified VSV-G containing unique blinding mutations that blind it to LDLR.
The G-Link CD3 adapter protein contains a VSV-G-binding region that binds to wild-type VSV-G, blocking its LDLR binding pocket.
Retargeting
The VSV-G glycoprotein is retargeted to the chosen receptor (CD3).
Cellective Targeting Adapter, covalently binds to modified VSV-G using SpyTag-SpyCatcher technology; the adapter contains an αCD3 scFv for retargeting that also activates T cells.
The G-Link CD3 adapter protein that binds wild-type VSV-G also contains an αCD3 scFv for retargeting that also activates T cells.
