K562-Fluc-Puro/CyCD20-Neo

K562-Fluc-Puro/CyCD20-Neo is a clonal population of the chronic myelogenous leukemia (CML) cell line K562 that has been engineered to stably express firefly luciferase and cynomolgus macaque CD20.

  • Species: Human
  • Cell Type: Chronic Myelogenous Leukemia
  • Endogenous Target Antigen: GPRC5D (low)
  • Transgene: Firefly luciferase (Fluc; Luc2), Cynomologu macaque CD20
  • Selection Gene: Neomycin, Puromycin
Description

The K562-Fluc-Puro/CyCD20-Neo cell line was generated by transducing Imanis K562-Fluc-Puro (CL171) cells with lentiviral vector encoding cynomolgus macaque CD20 and the neomycin resistance gene (LV-SFFV-CyCD20-PGK-Neo (LV072)). Transduced cells were subsequently selected using G418 followed by clonal selection using a methylcellulose based semi-solid medium.

This cell line has been tested for mycoplasma and no mycoplasma has been detected. The parental K562 cell line was authenticated by short tandem repeat (STR) profiling.

Characterization

Morphology

Low and high density cell photos (200x).

Luciferase Expression

The indicated number of cells were placed in wells of a 96-well plate. After the addition of 3 mg/mL d-luciferin, bioluminescence was immediately read using a TECAN plate reader.

CD20 Expression

K562-Fluc-Puro/CyCD20-Neo cells were stained with an anti-HuCD20/CyCD20 (blue) or isotype control (grey) antibody and analyzed by flow cytometry. (Note: the antibody used for analysis cross-reacts with both human and cynomolgus macaque CD20.)

Surface Receptor Profiling

K562-Fluc-Puro/CyCD20-Neo cells were stained with an anti-HuBCMA (A), anti-HuCD19 (B), anti-HuCD38 (C), or anti-HuGPRC5D (D) antibody and analyzed by flow cytometry.

Growth Conditions

Complete Growth Medium: Iscove’s Modified Dulbecco’s Medium (IMDM) supplemented with 15% fetal bovine serum (FBS) and 1X Penicillin/Streptomycin.

The addition of 6 µg/mL puromycin and 1 mg/mL G418 to the complete growth medium is recommended if maintaining the cells in culture for more than two weeks.

Grow cells in the indicated medium, subculturing as needed to maintain a density between 5 x 105 and 2 x 106 cells/mL. Passage cells by dilution in fresh medium with occasional complete media changes.

Usage Information

These cells are suitable for in vitro and in vivo experimentation.

The luciferase transgene facilitates noninvasive bioluminescent imaging of implanted cells in vivo and cell-based assays in vitro.

K562 cells form subcutaneous tumors and systemic disseminated tumors at multiple sites (liver, spleen, bone marrow) upon implantation into immunocompromised mice.

These cells can be amplified to generate additional frozen stocks and cryopreservation of low passage stocks in a designated cryopreservation medium is recommended.

These cells were generated by transduction with a self-inactivating Risk Group 3 (RG3) lentiviral vector. All work should be performed under appropriate biosafety conditions per institutional guidelines.
